~3H-TdR掺入法测定NHG-1脾淋巴细胞转化功能

本文报告用~3H-TdR掺入法测定本室的NC系正常裸小鼠和荷人脑胶质瘤裸小鼠(NHG-1)的脾淋巴细胞转化功能。结果提示:正常的纯合子(nu/nu)T细胞缺乏,B细胞功能均正常;杂合子(nu/ )的T和B细胞功能均正常;荷瘤鼠(NHG-1,30代)除了T细胞缺乏,B细胞功能亦受到抑制。     

中西结合治疗椎动脉型颈椎病62例临床观察

1 临床资料 本组62例中,男38例,女24例;年龄最大67岁,最小21岁,平均36岁;病程最长8 a,最短2个月,平均15个月.全部病例均有眩晕症状,常在颈部旋转或屈伸时出现或加重,伴颈枕部不适或疼痛,颈肌僵硬.伴耳鸣32例,伴恶心或呕吐23例,伴上肢麻木感15例.     

Benign giant cell tumor of bone with osteosarcomatous transformation (”dedifferentiated” primary malignant GCT): report of two cases

 It is not uncommon for sarcomatous transformation of giant cell tumor (GCT) of bone to occur after radiation, but rarely does malignant transformation occur spontaneously, with less than 15 cases reported up to 1995. Only four of these cases have been documented in detail. We report two additional cases of GCT of bone spontaneously transforming or ”dedifferentiating” into osteosarcoma without radiation therapy. The first case is absolutely unique and most interesting in that the dedifferentiation process occurred in one of multiple GCT lung metastases 6 years after successful eradication of a primary tibial tumor. The right lung was resected due to development of a large tumor, and at pathologic examination, demonstrated several small nodules of conventional GCT and a much larger, 14-cm mass composed of a mixture of GCT and high-grade osteosarcoma. The second case involved a physician, who had a large tumor in the sacrum with vague symptoms for 8 years. Open biopsy revealed conventional, benign GCT of bone with a secondary aneurysmal bone cyst. Complete curettage 2 weeks later revealed, in addition to areas of conventional, benign GCT a second component of very high grade osteosarcoma. Both patients died less than 1.5 years from diagnosis. This report of osteosarcomatous transformation of a conventional GCT of bone strengthens the theory that there is a mesenchymal cell line in GCT that may spontaneously tansform to sarcoma.     

Effect of “Re Du Qing” on the activation,proliferation and membrane fluidity of lymphocytes

Summary Effects of Chinese Medicinal Preparation “Re Du Qing” (RDQ) on the activation, proliferation and membrane fluidity of T lymphocytes from human peripheral blood Were studied by means of³H-TdR incorporation and DPH fluorescence polarization. The results showed that “RDQ” can:1) significantly inhibit the activation of T lymphocytes; 2) restrain the proliferation of activated T lymphoblasts in the presence of exogenous interleukin-2 (IL-2); and 3) increase the membrane fluidity of T lymphocytes and antagonize the decreased fluidity of lymphocyte membrane mediated by Con A or PHA. The functional abnormalities of T lymphocytes in some autoimmune diseases such as arthritis and the usefulness of RDQ in the treatment of these diseases were also discussed.     

The Construction fo Hybrid Plasmid pBR322:RSF1010

The plasmids pBR322 and RSF1010,extracted from E.coli HB802 and E.coliC600 respectively,were treated with restrictive endonuclease EcoRI andligated with T4 DNA Iigase.C600 strains were then transformed with thehybrid plasmid(pSMMl),and thus253transformants were obtained.Thefigures of gel electrophoresis and electromicrograph proved that pSMMlwas a pBR322:RSF1010 composite hybrid.In E.coli C600 pSMMl obtainedresistance against ampieilline(Ap),tetracycline(Tc)and streptomycin(Sm).The orientation of pBR322 and RSF1010 in hybrid pSMMl and the failureof introduction of this hybrid into P.putida AC10 were discussed.     

A Drosophila melanogaster hobo-white+ vector mediates low frequency gene transfer in D. virilis with full interspecific white+ complementation

Transformation of a Drosophila virilis white mutant host strain was attempted using a hobo vector containing the D. melanogaster mini-white⁺ cassette (H[w⁺, hawN]) and an unmodified or heat shock regulated hobo transposase helper. Two transformant lines were recovered with the unmodified helper (HFL1), one containing only the white⁺ marked vector, and a sibling line containing the vector as well as an HFL1 helper integration. An approximate total transformation frequency of 1% is deduced. A high frequency of wing and eye morphology mutants were also observed, suggesting that hobo may have mobilized a related element in D. virilis. The data reaffirms a relatively low transformation vector activity for the hobo transposon in D. virilis; however, nearly full interspecific expression of the white⁺ marker supports its possible function in other species as well.     

煤焦沥青烟雾提取物的细胞毒性和遗传毒性研究

采用人外周血淋巴细胞非程序DNA合成(UDS)试验和人胚肺成纤维细胞转化试验,测试了煤焦沥青烟雾提取物(ECTPF)对人体细胞的细胞毒性和遗传毒性。UDS试验结果表明,ECTPF可使淋巴细胞UDS值明显增加,并有剂量一反应关系。引起半数淋巴细胞死亡的浓度(LC50)为33.8μg/ml。细胞转化试验表明,ECTPF能诱发人胚肺成纤维细胞明显的形态学转化,且转化细胞具有部分恶性转化细胞的特性。引起半数人胚肺成纤维细胞生长抑制的浓度为41.3μg/ml。实验结果提示,ECTPF是一种具有细胞毒性和遗传毒性的物质。     

低温手术过程多维传热有限元分析

目的 通过有限元分析，给出低温手术过程组织三维瞬态温度场；考察人体血液灌注和代谢产热对其影响，为优化设计冷刀提供参考。方法 充分考虑组织物性和内热源的非线性，使用基于有限容积法的大型商业有限元软件Ansys求解描述低温手术过程组织传热问题的经典生物传热方程。结果 考虑血液灌注和代谢产热后，组织局部降／复温速率降低、所经历的最低温度绝对数值减少；对应时刻，组织内局部温度梯度增加；相同手术方案下，组织内冻结区域减小，且冰球生长过程减慢。结论 人体血液灌注和代谢产热对低温手术过程影响显著，体外实验由于无法模拟血液灌注和代谢产热效果而对冷刀的优化设计作用有限，必须进行在体实验以优化冷刀及其相应的低温手术方案。     

门静脉海绵样变的手术治疗

目的 探讨门静脉海绵样变的手术治疗方式。方法 对9例门静脉海绵样变患者的14次手术方式进行回顾分析。结果 1例患者剖腹探查无法手术，其余13次手术中，分流手术5次，断流手术8次，对于再出血时间的对比分析，断流手术发生再出血时间长，并与断流的彻底性有关。结论 对于门静脉海绵样变的手术方式以彻底的断流术为佳，最好行贲门周围血管离断 食道下段 胃底切除再吻合术。     

三种淫羊藿的总黄酮分极提取物对小鼠免疫功能的影响

观察了巫山淫羊藿、柔毛淫羊藿和粗毛淫羊获的总黄酮正丁醇提取物（代号Ａ１、Ａ２、Ａ３）和巫山淫羊藿总黄酮乙酸乙酯提取物（Ｂ１）对正常小鼠免疫功能的影响。结果；Ａ１可以明显促进脾脏抗体形成细胞的功能（Ｐ＜０．０５），Ａ２、Ａ３虽也有一定的促进作用，但无统计学意义；４种提取物在体外实验中均没有协同ＣｏｎＡ诱导牌细胞增殖作用，只有提取物浓度在１００μｇ／ｍｌ时，有一定抑制作用。４种提取物对２４小时培养脾细胞存活率无明显影响。
